No, messages can't be unsent. No, they got the message immediately after you sent it. Investigating the listing and the seller's feedback is something you should have done before you bid. The seller has probably already started a Cancellation and have put you on their Blocked Bidder list, so you are probably SOL.
The Best Offer is a commitment to buy, the same as making a bid on an auction item or purchasing an item at full price. Make sure you really want the item, because if you don't pay, the seller can file an unpaid item strike against you. Too many unpaid item strikes and your eBay account can be shut down.
The seller has to wait until the buyer pays if the seller accepts an eBay offer. The item will no longer be visible as available for others to purchase and will appear on the seller's Awaiting Payment list.

I generally shoot for around 65%-70% of their asking price as a first offer.
But lowballers, I just laugh at the $3 or $5 offer and decline it as opposed to blocking them. I definitely wouldn't block unless it's someone who you really would feel could harm your business by either harrassment or being vengeful. Counter low offers, most low ballers will move on.
If they try to interact with your listings, they will be notified that they've been blocked. Go to their profile and find the icon in the upper right hand corner of the screen. From here you can click Block User, which will automatically unfollow you from this person and vice versa.
Yes, Poshmark is a reputable website. Buyers purchase actual clothes, and sellers receive actual payouts. That being said, while the site itself is perfectly legitimate, there is no guarantee that the users buying and selling on it are legitimate.
